Solid phase microextraction (SPME) is a relatively new and sensitive solvent-free sample preparation technique. Based on the principle of adsorption/absorption and desorption, SPME uses a coated fiber to concentrate volatile and semi-volatile compounds from a sample.
SPME uses a fiber coated with polyermers and sorbents. The coated fiber is placed in a protective needle and attached to a SPME holder. When the fiber is exposed to a sample, the anlytes of interest partition from the sample matrix into the stationary phase until an equilibrium is established. The fiber’s coating extracts target compounds from the sample either by absorption or adsorption. After a predefined extraction time, the fiber is removed and inserted directly into a chromatographic instrument, usually gas chromatography(GC) or high-performance liquid chromatography(HPLC), for desorption and analysis.
SPME combines the analyte sampling, isolation and enrichment into one step, make it easy to automate the procedures.
