Why Perform Single-Cell Sequencing?


Traditional sequencing is conducted on the tissue level, and the results are averages of multiple cells' genetic information, making it impossible to pinpoint the genetic information of individual cells. In reality, there is heterogeneity between cells, even within tumor tissues. The genetic information at the genome and transcriptome levels differs among cells at the tumor's center, surrounding cells, and cells that have metastasized to distant locations. Single-cell sequencing involves sequencing DNA or RNA at the single-cell level to analyze individual cells' gene structures and expressions, achieving higher resolution in gene expression analysis.


Currently, single-cell sequencing mainly includes single-cell genome sequencing, single-cell transcriptome sequencing, and single-cell epigenome sequencing.


Early single-cell sequencing processes first involved isolating single cells through dilution, flow sorting, or microdissection, followed by micro-library construction. This process was complex, had low throughput, and was costly. Today's mainstream high-throughput single-cell capture sequencing uses barcodes to label individual cells, allowing for the mixing of multiple cells for library sequencing. During subsequent data analysis, molecular information marked with the same barcode is considered to be from the same cell.
